This is a new PC so I don't think it can be a hardware problem. Everything else is fine and so was iTunes a few hours ago

I had a similar experience with a different program earlier this year. It turned out to be a computer that was failing. In spite of new hard drive, and new RAM - the motherboard itself was breaking down. The computer was 9 1/2 years old - replaced it and the program worked perfectly again. That new hard drive I bought is now a second drive in the new computer. The RAM was a waste of money.
I usually wait a while before installing new iOS or any new program, until the wrinkles get ironed out. I am surprised they didn't find the iOS10 issues in the extensive beta program.
The free program Media Monkey is a good backup program if iTunes is acting up, it will at least let you into your devices.
