I like mine set to use the right thumb, right index and left thumb. That lets me unlock the iPhone no mater how I am holding it.

I prefer to have the option to use either of two of my fingers. If you want to use only one, go to Settings>Touch ID & Passcode and swipe to remove the fingerprint you no longer wish to use. Here's more info on the feature from Apple:
