Bingo!

I re-converted the massive Quicktime movie into .mp4 using Handbrake
and iPhone presets which gave a 200MB file and 960 x 540px.

I tried the File Transfer using iTunes - it seemed to transfer but I
couldn't find the file on the phone.

I then dropped it on the iTunes icon - it appeared in the Library,
appropriately under Movies/Home Videos.

In the Sync window I checked the box next to my video and synced.

The video appeared in the Videos app, under a new tab called "Home
Videos". The quality on the small screen at the reduced resolution was
more than adequate.

Flushed with success I the loaded a small .mov video to check the process.
